Retinoids are arguably the best-studied anti-aging and acne-treating ingredients in all of dermatology. They are a family of compounds related to vitamin A, and most of them work in a very similar way: they encourage your skin cells to turn over more quickly, calm the enzymes (called MMPs) that break down collagen, and switch on the signals that build new collagen. Together, that explains why they help with fine lines, texture, uneven skin tone, breakouts and dark spots.
What varies a lot between them is how strong they are, how well they are tolerated, and how much human research actually backs them up. Some, like prescription tretinoin, have decades of human studies behind them and are considered the gold standard. Others, like retinyl palmitate, one of the mildest forms, have largely lab or animal data, and a 48-week human study of a related ester (retinyl propionate) failed to beat placebo. So while they share a mechanism of action, the evidence stacks up differently for all of them.
To complicate things, all retinoids share a common family of skin receptors (RARs), but they differ in:
Potency — how active they are once they reach your skin
Conversion steps — esters must be converted to retinol, then to retinal, then to retinoic acid; each step reduces strength, but increases tolerability.
Receptor selectivity — newer retinoids target specific RAR subtypes for better tolerability
Delivery systems — encapsulated and micropump formulations reduce irritation
Systemic vs topical — oral isotretinoin works throughout the body; topical retinoids stay on the skin
It is important to understand these key differences so you can make an educated decision on which type to start with on your retinoic acid journey. The rest of this post walks through each retinoid in the family, from mildest to strongest, with the pros, cons and key points of each.

1. Retinyl Palmitate, Retinyl Acetate, Retinyl Propionate (Retinyl Esters)
Strength: Mildest in the family
How they work: These are esters of vitamin A. Before they can do anything, your skin has to convert them in three steps. First to retinol, then to retinal, then to retinoic acid, and only the final form binds to retinoid receptors. Because of those multiple conversion steps, very little active ingredient actually reaches your skin's target cells.
Pros:
Very well tolerated, with low risk of redness, peeling or dryness
Stable in light and air, so they hold up well in skincare formulas
Good starting point for sensitive skin or people nervous about stronger retinoids
Cons:
Weakest of the vitamin A family because of those three conversion steps
Most of the evidence comes from lab and animal studies rather than human trials
A 48-week study of 0.15% retinyl propionate alone failed to beat placebo for photoaging
Relevant research:
Retinyl palmitate has shown anti-photoaging activity in UVB-irradiated mouse models, with improved collagen, elastin and hyaluronic acid synthesis. Human skin penetration studies show it absorbs less effectively than retinol or retinoic acid.
Retinyl propionate combined with another ingredient beat 0.1% retinol for deep wrinkles in one 16-week trial, but the same ester alone didn't outperform placebo in a 48-week study.
Retinyl acetate is another very stable cosmetic ester, but there are no published clinical trials of it used alone as an anti-aging ingredient.
Best for: Someone with very sensitive skin, or anyone using retinoids for the first time, who wants a very gentle entry point.
2. Retinol
Strength: Weak to moderate; roughly 20 times less potent than tretinoin
How it works: Retinol skips a conversion step compared to retinyl esters. It still needs one enzymatic conversion to retinal and another to retinoic acid, but it reaches active form more efficiently than esters.
Pros:
Widely available over the counter
Strong human evidence for reducing sun-damage enzymes and boosting collagen
Among the best-tolerated retinoids in the family
Many modern formulations use encapsulation, which releases the retinol slowly and further reduces irritation
Cons:
Much weaker than prescription options, so you will see slower or less dramatic results
Can still cause redness, dryness or peeling, especially in the first few weeks
Quality varies widely between brands
Relevant research:
Retinol inhibits UV-induced MMPs and stimulates collagen synthesis in photoaged human skin, with one study showing 1% topical retinol on aged skin for 7 days increased dermal fibroblast activity, reduced MMP expression and increased collagen.
A 2021 randomized controlled trial showed topical 0.3% retinol for 12 weeks produced statistically significant reductions in wrinkle depth and improved skin texture, with less redness than tretinoin.
It up-regulates TGF-β/CTGF (collagen-building signals) and down-regulates CCN1, a molecule that normally holds collagen production back.
Best for: Most adults wanting an anti-aging or skin-smoothing routine without prescription strength; a sensible first retinoid for most people.
3. Retinaldehyde (Retinal)
Strength: Moderate; stronger than retinol, gentler than tretinoin
How it works: Retinal is only one conversion step away from active retinoic acid. It is converted in the skin by differentiating keratinocytes, which means the conversion is more controlled and tends to cause less irritation than directly applying tretinoic acid.
Pros:
More potent than retinol for fine lines and pigmentation
Less irritating than tretinoin
Newer encapsulated formulations deliver retinal deeper into the skin with minimal irritation, even in sensitive or rosacea-prone skin
Cons:
Generally only available over the counter at higher price points
Still a "cosmeceutical," not FDA-approved to treat any condition
Slightly less robust human data than tretinoin
Relevant research:
Human randomized trials show 0.05% retinaldehyde improves both fine and deep wrinkles vs vehicle, and with less irritation than tretinoin.
A 12-week study of encapsulated retinal showed improvement in mild to moderate photoaging with minimal irritation, even in sensitive skin.
Best for: People who have outgrown retinol but don't want (or can't get) a prescription; also a good option for those who find tretinoin too harsh.
4. Hydroxypinacolone Retinoate (HPR / Granactive Retinoid)
Strength: Moderate to strong (direct receptor activation, no conversion needed)
How it works: Unlike other retinyl esters (remember these are the weakest ones), HPR does not need to convert to retinoic acid. It binds directly to retinoid receptors (particularly RXRs, and in newer data, RARα). This skips the multiple conversion steps that weaken other esters, which is why it's often marketed as a "next-generation retinoid."
Pros:
Directly activates retinoid receptors without conversion, so it behaves more like an active retinoid than a precursor
Less inflammatory (lower IL-1α) and more stable than tretinoin in skin models
In a UVR-induced photoaging model, 9-cis HPR promoted collagen and elastin production while reducing erythema and desquamation (peeling)
Better tolerability than retinol and tretinoin in combination studies
Cons:
Still considered a cosmetic-grade retinyl ester (not FDA-approved to treat any condition)
Human data remain limited compared to retinol or tretinoin
Most clinical research uses HPR in combination with retinyl propionate, not alone
Relevant research:
HPR significantly increased pro-collagen production in skin models with less IL-1α than tretinoin.
9-cis HPR in a photoaging mouse model restored fibroblast and melanocyte proportions, up-regulated collagen-related genes and selectively activated RARα and RXRα.
Combined HPR and retinyl propionate improved wrinkles, skin elasticity and smoothness in human subjects with similar efficacy to retinol but better tolerability.
Best for: Those who want a more potent OTC retinoid than retinol or retinaldehyde but prefer the tolerability profile of esters. Also a good choice for people who want retinoid benefits with less irritation. If the lack of human data is concerning, skip this and go for a retinol.
5. Adapalene (Differin, La Roche-Posay Effaclar)
Strength: Moderate; prescription at 0.3%, over the counter at 0.1%
How it works: Adapalene is a synthetic third-generation retinoid that selectively targets RARβ and RARγ receptors. This receptor selectivity is why it tends to be gentler than older retinoids.
Pros:
Excellent tolerability, with significantly less irritation than tretinoin or tazarotene
Photostable, so it can be used in the morning and combined with benzoyl peroxide
Available over the counter at 0.1%
FDA-approved and gold standard for mild to moderate acne
Cons:
Less evidence for anti-aging specifically compared to tretinoin
Can still cause dryness or irritation, especially in the first few weeks
Pregnancy category C, so avoid if planning pregnancy
Relevant research:
Multiple trials show adapalene 0.1% is as effective as tretinoin 0.025% for acne with significantly less irritation.
Systematic reviews consistently place adapalene at the top of the tolerability list among prescription retinoids.
Best for: Acne-prone skin, sensitive skin, teenagers, or anyone who wants a well-tolerated prescription-strength retinoid without the redness of tretinoin.
6. Tretinoin (Retin-A, Altreno, Atralin, others)
Strength: Strong; the gold-standard prescription retinoid
How it works: Tretinoin is the active form of retinoic acid, so it doesn't need any conversion in the skin. It binds directly to RARα, RARβ and RARγ.
Pros:
The most studied anti-aging ingredient in dermatology
FDA-approved for photoaging and acne
Griffiths' landmark study showed ~80% increase in type I collagen in sun-damaged human skin
Reduces UV-induced MMP activity by 70 to 80% in human skin
Cons:
Highest irritation risk among retinoids, with redness, peeling, dryness, especially in the first few months
Not photostable (apply at night)
Pregnancy category C - stop use in pregnancy.
Takes 3 to 6 months to see anti-aging benefits
Relevant research:
Griffiths et al., New England Journal of Medicine: 0.1% tretinoin cream produced ~80% increase in type I collagen formation in photodamaged skin over 10 to 12 months.
Multiple RCTs show improvement in fine wrinkles, mottled hyperpigmentation and skin texture vs placebo.
Newer microsphere, micropump and foam formulations (Altreno, Atralin, Retin-A Micro) deliver the same active ingredient with substantially less irritation.
Best for: Adults with clear photoaging (fine lines, texture, pigmentation), stubborn acne, or those wanting the strongest available topical anti-aging effect with the most clinical evidence.
7. Topical Isotretinoin (Isotrex)
Strength: Strong; not commonly available in the US, available through certain compounding pharmacy companies, typically with a online visit with dermatologist
How it works: Isotretinoin is a non-selective RAR agonist, like tretinoin, but appears to have more anti-inflammatory effects.
Pros:
Similar anti-aging and acne efficacy to tretinoin, but with fewer side effects like stinging and peeling in direct comparisons
In a recent network meta-analysis, it ranked first of all anti-aging topicals for improving fine lines
Can be helpful for people who can't tolerate tretinoin
Cons:
Not FDA-approved in the US for photoaging; limited availability
Still a retinoid, so same pregnancy precautions as tretinoin
Relevant research:
Human trials show 0.05% to 0.1% topical isotretinoin improves fine wrinkles, texture and sun spots.
A 2025 network meta-analysis ranked topical isotretinoin first for fine wrinkle improvement vs placebo.
Best for: Those wanting a strongly evidenced prep, without the irritation of Tretinoin.
8. Tazarotene (Tazorac, Fabior)
Strength: Very strong; the most potent topical retinoid available
How it works: A third-generation retinoid applied as a prodrug (tazarotene) that the skin converts to its active form (tazarotenic acid), which has particularly high affinity for RARβ and RARγ.
Pros:
Most potent topical retinoid on the market
Beats tretinoin for coarse (deep) wrinkles and works faster; one network meta-analysis showed odds ratio 3.95 vs tretinoin
FDA-approved for acne, photoaging (0.1% cream) and psoriasis
Available as a foam (Fabior) which improves tolerability
Cons:
Highest irritation risk, with burning and peeling in the first week very common
Pregnancy category X, so absolutely contraindicated in pregnancy or planning pregnancy
Often too strong for sensitive skin without a build-up period
Relevant research:
Tazarotene 0.1% cream vs tretinoin 0.05% cream: comparable wrinkle improvement with faster onset at 12 weeks.
Network meta-analysis: superior to tretinoin for coarse wrinkles (OR 3.95, p=0.009).
Best for: Patients with significant coarse wrinkles or thick sun-damaged skin who can tolerate the irritation, or those with psoriasis; always start low and build up gradually.
9. Trifarotene (Aklief)
Strength: Very strong; the newest prescription retinoid (approved 2019)
How it works: A fourth-generation retinoid that selectively targets the RARγ receptor, the dominant RAR subtype in the epidermis. This selectivity was designed to improve tolerability and specificity.
Pros:
FDA-approved for acne in patients 9 years and older, including truncal acne (chest and back), uniquely approved for body acne
Emerging data suggest it may improve acne scarring and post-inflammatory pigmentation earlier than older retinoids
Approved across four phase III trials (PERFECT 1 & 2, DUAL, START, LEAP)
Cons:
Can still cause irritation, similar to other strong retinoids
Head-to-head tolerability data vs tazarotene and tretinoin are still building
Pregnancy category, same precautions as other retinoids
Relevant research:
PERFECT 1 & 2 trials: significant reduction in inflammatory and comedonal acne lesions on the face and trunk at week 12.
Some trial data show improvements in acne scarring and hyperpigmentation as early as week 2, though more study is needed.
Best for: Teens and adults with acne on the face and trunk, or those who haven't responded well to older retinoids.
10. Oral Isotretinoin (Accutane, Amnesteem, Claravis, others)
Strength: The strongest, a systemic (whole-body) treatment
How it works: Taken by mouth, isotretinoin reduces sebaceous (oil) gland size and output, normalizes keratinization, and has significant anti-inflammatory effects. It is the most powerful acne medication available.
Pros:
Most effective acne treatment available, and can lead to long-term and sometimes permanent remission in severe cystic acne
The only acne medication that can permanently alter the course of the disease for many patients
Cons:
Multiple systemic side effects: severe dryness of skin, lips and eyes; potential mood changes; liver and lipid changes; bone effects, joint pain, Meibomian gland dysfunction
Absolutely contraindicated in pregnancy, as it causes serious birth defects. Requires enrollment in the iPLEDGE program in the US
Requires regular blood monitoring
Strict sun precautions
Months of treatment with strict follow-up
Relevant research:
Decades of evidence support its use for severe nodulocystic acne resistant to other treatments.
Newer lower-dose regimens are gaining support with similar outcomes and fewer side effects.(Microdosing)
Best for: Severe, treatment-resistant, or scarring acne, always under the care of a dermatologist.
Bonus: Bakuchiol, The Retinol Alternative (Not a Retinoid)
Strength: Moderate; works like retinol but is a plant compound, not vitamin A
How it works: Bakuchiol comes from the Psoralea corylifolia plant. It does not bind to retinoid receptors directly, but it activates many of the same collagen- and MMP-related genes as retinol. In lab studies, it reduces PGE2 (a molecule that promotes MMP-1 and inhibits collagen) and upregulates collagen type VII, sometimes more effectively than retinol itself.
Pros:
Comparable efficacy to retinol in a 12-week randomized trial, with significantly less irritation
Safe in pregnancy and breastfeeding (no retinoid risk)
Photostable, so it can be used day and night
Good for people who can't tolerate any retinoid, or who are pregnant
Cons:
Less human data than retinol, and far less than prescription retinoids
Not FDA-approved to treat any specific condition
Sourcing and purity vary widely between brands
Relevant research:
Gehring 2019 (British Journal of Dermatology): 12-week trial comparing 0.5% bakuchiol cream with 0.5% retinol cream. Bakuchiol matched retinol for wrinkle and pigmentation improvement with significantly better tolerability.
Best for: Pregnant or breastfeeding patients, people with very sensitive skin or rosacea, or anyone wanting a plant-based alternative to retinol.

Regardless of which you choose, remember the universal rules:
Start low and go slow. 2 to 3 nights per week at first, then build up to daily use.
Use at night. Most retinoids are not photostable (exceptions: adapalene, bakuchiol, HPR).
Always use sunscreen. All retinoids increase sun sensitivity.
Expect retinoid adjustment. Some redness, dryness or peeling in the first few weeks is normal.
Be patient. Visible anti-aging results typically take 3 to 6 months.
And if you are pregnant, planning pregnancy, or breastfeeding, avoid all prescription retinoids. Tazarotene, topical isotretinoin and oral isotretinoin are absolutely contraindicated, and talk with your dermatologist before using any retinoid at all.
